In the dementia network, this interactome is connected to presenilin (Figure 5) whose mutation underlies the majority of familial Alzheimer’s disease cases[56-58] and whose role in autophagy has been shown to be central[59], presenilin 1 being essential for lysosome acidification, and proteolysis during autophagy[60]. This evidence concerns the gene PSEN1 and early-onset autosomal dominant Alzheimer disease.
