Since α-latrotoxin induces synaptic vesicle release and LPHN3 mutations are linked to cortical function through their association with ADHD, we chose to investigate two hypotheses regarding the role of presynaptic LPHN3 at cortical synapses: (1) LPHN3 modulates presynaptic vesicle release; and (2) LPHN3 regulates synapse development. This evidence concerns the gene ADGRL3 and attention deficit-hyperactivity disorder.
