The concentrations of total PYY observed with 300 mg BID of GSK263 in our T2D studies (up to 50 pM in Study 1 and 100 pM in Study 2; presumed to be mainly PYY3–36) should have reduced food intake based on the reported effects of acute PYY3–36 infusion [5], [23], yet we did not observe changes in the self-reported hunger, satiety or craving parameters, or estimations of caloric intake. This evidence concerns the gene PYY3 and type 2 diabetes mellitus.
