TNFRSF8 and anaplastic large cell lymphoma: Brentuximab vedotin (ADCETRIS®) is an antibody-drug conjugate composed of a CD30-targeted chimeric monoclonal antibody (cAC10) covalently linked, via a protease-cleavable linker, to the microtubule-disrupting agent monomethyl auristatin E. Results from phase 2 pivotal trials of single-agent brentuximab vedotin (1.8 mg/kg) demonstrated an objective response rate (ORR) of 75% (34% complete remission [CR]) in Hodgkin lymphoma (HL) patients and 86% (57% CR) in systemic anaplastic large cell lymphoma (ALCL) patients[1,2].
