Disruption or misregulation of the Shh pathway results in various developmental abnormalities including holoprosencephaly, Pallister–Hall syndrome, Gorlin syndrome, Greig cephalopolysyndactyly, Rubinstein–Taybi syndrome and different types of cancer (basal cell carcinoma, medulloblastoma, glioma, breast, pancreatic, prostate cancers and more). This evidence concerns the gene SHH and glioma.
