An exciting study published recently in Nature [14], together with an earlier one [15], has verified a new class of small molecule inhibitors, XAV939, which could block Wnt signaling in colon cancer cell lines by binding to tankyrase (TNKS) catalytic poly-ADP-ribose polymerase (PARP) domain, and then resulted in dramatic stabilization of the Axin protein, thereby lead to increased β-catenin destruction. This evidence concerns the gene TNKS and colonic neoplasm.
