Consistent with the clamp data, phosphorylation of the serine/threonine kinase, AKT, is increased in livers isolated from insulin-stimulated KLF15-/- versus WT mice after 8 weeks of HFD; however, these mice also exhibit increased hepatic expression and phosphorylation of UPR genes compared to WT littermates, providing evidence for uncoupling of ER stress and insulin resistance (Figure 2A).
