The nucleolar RNA binding shuttling protein nucleolin appears to be a common target of virus action, having previously been shown to mislocalise to the cytoplasm upon infection with HRV14 [21], poliovirus [29], [40], feline calicivirus [41], and even herpes simplex virus 1 (HSV-1); here we confirm similar results for HRV16 infection, and demonstrate mislocalisation from the nucleolus to the nucleoplasm in cells expressing active GFP-3C protease (see Figure 3C), concomitant with cleavage of the protein, either directly by 3C or in a 3C protease-dependent manner (see Figure 3).
