The most direct evidence to date that interfering with TLR4 might have therapeutic potential in RA was shown by Abdollahi-Roodsaz et al., where inhibition of TLR4 signaling with a specific TLR4 antagonist prevented the inflammatory feedback loop in a mouse model of autoimmune destructive arthritis, and suppressed both clinical and histologic characteristics of arthritis [13].
