MTOR and tuberous sclerosis: The landmark Multicenter International LAM Efficacy of Sirolimus (MILES) Trial, a randomized, placebo controlled trial in 89 women with sporadic or TSC-associated LAM, demonstrated that Rapamycin (an allosteric mTOR inhibitor) stabilized lung function during one year of therapy; discontinuation of therapy resulted in a rate of lung function decline similar to untreated patients [12].
