While the treatment with CCI-779 alone, used in a wide range of concentrations (1–2,000 nM), exerts only a minor cytostatic effect on melanoma cells (data not shown), the co-treatment with cisplatin affords a remarkable synergistic effect on cell death produced by cisplatin alone, in terms of both cell detachment and caspase-3/-7 activity (Fig. 10B and C).
