The mechanisms responsible for activation of the PI3K/AKT pathway in human cancers are diverse and include dysregulation of growth factor receptor and integrin signalling, activating RAS mutations, activating mutations or gene amplification of the gene encoding the p110α catalytic subunit of PI3K (PI3KCA), inactivating mutations in the phosphatase and tensin homolog (PTEN) tumor suppressor gene or in the gene encoding the p85 regulatory subunit of PI3K (PI3KRA).
