When testing the compounds in the CFC assay with haematopoietic progenitor cells (CD34+) derived from Jak2V617F-positive MPN patients, we found that in general Jak inhibitors preferentially suppressed growth of erythroid colonies, whereas Aurora kinase inhibitors (AT9283, VX680, AZD1152, Danusertib and SNS314) preferentially blocked myeloid colony growth (Fig. 4 and Figure S7). The gene discussed is CD34; the disease is myeloproliferative neoplasm.
