Our studies indicate that ADAM17 is a target gene of miR-145, and several lines of evidence support the direct interaction between miR-145 and ADAM17: i) the 3′-UTR of ADAM17 contains a binding site for miR-145 with significant seed match; ii) miR-145 suppresses the activity of a luciferase reporter with the 3′-UTR of ADAM17 mRNA; iii) miR-145 represses ADAM17 expression at protein level; (4) our previous studies have shown that ADAM17 knockdown by siRNAs resulted in a decreased glioma cell proliferation, migration and invasiveness.
