To determine whether Elesclomol treatment was linked to ROS produced during the process of melanin synthesis [14], we also treated the amelanotic metastatic melanoma cell line C32 with Elesclomol. C32 cells lack functional tyrosinase protein and therefore do not produce melanin in response to ultraviolet irradiation. As shown in Fig. 1, Elesclomol showed similar cytotoxicity in C32 cells as it did in the pigmented WM1158 cells, which indicates that non-melanosomal-containing organelles are important for Elesclomol-mediated increase in ROS.
