EGFR may be another EGCG-direct target that through inhibition of its downstream signalers (Akt, ERK1/2 and mTOR) is able to down-regulate FASN expression at two different levels: 1, at the transcriptional level through the sterol response element-binding proteins 1c (SREBP-1c), the FASN-transcription factor mediated by PI3K/Akt and MAPK/ERK1/2 pathways [39]; 2, at the translational level, through Akt-mTOR-signaling and its downstream effectors, eIF4G and S6K (reviewed in ref [40]) as seen in breast cancer [41] and in human hepatoma cells [42]. The gene discussed is FASN; the disease is hepatocellular carcinoma.
