To determine the ability of miR-34a to regulate Notch pathway players in NPC, we transduced NPC with FIVneorU634a or FIVneorU6cntrl vectors, selected the transduced NPC by culturing in G418, and assessed mRNA levels of Numbl, Notch1, Cbf1, NeuroD1, and Mash1. Interestingly we found that transcript levels of Notch1 and Cbf1 were elevated in the context of miR-34a over-expression (Fig 3b). This evidence concerns the gene ASCL1 and nasopharyngeal carcinoma.
