PPARG and osteosarcoma: For our subsequent studies we used the human U2OS osteosarcoma cell line, since these cells (i) are easy to handle and transfect, (ii) express a robust transcriptional response upon introduction of PPARγ, both on the level of reporter genes [36], [37] and endogenous target genes (data not shown), and (iii) may be growth inhibited by activation of PPARγ, a phenomenon also observed in multiple other cell lines [33], [38] which will support lethality rescue screening.
