A more recent study shows p62-based selective autophagy is involved in myeloid cell line differentiation via degrading PML–RARα [a fusion protein of promyelocytic leukemia (PML) and the retinoic acid receptor-α (RARα)], while inhibition of autophagy by shRNA targeting essential autophagy genes (ATG1 and 5) and pharmacological inhibition with 3-MA blocked both all-trans retinoic acid (ATRA)-induced PML–RARα degradation and consequently myeloid cell differentiation. This evidence concerns the gene RARA and acute promyelocytic leukemia.
