A TGFβ/FOXO/Akt signalling axis was found to be responsible for imatinib resistance in a CML stem cell population and it was shown that pre-treatment of CML stem cells with the TGFβ receptor inhibitor LY364947 efficiently activated Akt and suppressed FOXO-induced cell cycle arrest and apoptosis [17]. The gene discussed is TGFB1; the disease is chronic myelogenous leukemia, BCR-ABL1 positive.
