Interestingly, inhibition of mTORC1 by the rapamycin analog RAD001 was shown to potentiate the effect of 1,25-VD to induce growth arrest and differentiation in AML cells, which prompted the authors to propose the strategy of concomitant administration of both 1,25-VD and an mTOR inhibitor in treatment of AML [53]. The gene discussed is MTOR; the disease is acute myeloid leukemia.
