This study investigated a Southwestern American Indian tribe by genome-wide linkage analysis and reported linkage to marker D4S1645, which is less than 300 kb upstream of LPHN3. Another independent study utilized two-point linkage analysis in families with alcohol dependence to identify a nominally significant locus at D4S244 (Reich et al. 1998), which is ~5 Mb downstream of LPHN3. While its distance from LPHN3 makes this latter report less compelling, it nonetheless suggests that there is a locus on chromosome 4p related to alcohol dependence (Uhl 2004a, b). The gene discussed is ADGRL3; the disease is alcohol dependence.
