TP53 and cancer: The transcription factors p53, p63 and p73 play an important role in cancer prevention, development and longevity.1–4 The three proteins share a similar domain organization (Fig. 1a), being composed of structured DNA-binding domain (DBD) and tetramerization domain flanked by intrinsically disordered regions.2,7,8 The phylogenetically more ancient p63 and p73 proteins have an extended C-terminal region that is absent in p53.
