In fact, when proteasome activity was blocked proteins were found into cytosol at the same strength of the sc-siRNA (more evident at 20 μM drug concentration) confirming that the BAG1 expression maintains them away from the proteasome, particularly for MCL1 (by densitometric data: 3 fold recovery increase for BCL-2 for BCS, 6 fold increase for MCL-1, 2-fold for BCL-XL, Figure 6A), suggesting that its over-expression in AML is consistent with a low degradation rate of those proteins. This evidence concerns the gene BCL2L1 and acute myeloid leukemia.
