Subsequently, in a meta-analysis combining eight independent studies and comprising a total of 13,659 Europeans two common non-synonymous variants, rs6232, encoding N221D, and rs6235, encoding the Q665E-S690T pair, within PCSK1 were reported to associate with an increased risk of obesity with an OR of 1.34 and 1.22, respectively (the G-allele of rs6232: p = 7.27 * 10−8 and the C-allele rs6235: p = 2.31 * 10−12) [9]. Here, PCSK1 is linked to obesity due to melanocortin 4 receptor deficiency.
