While we did not find increased DNA methylation in any of our 15 DNA hypermethylation loci between high-grade or low-grade AAH, in an unsupervised analysis we identified a small group of five AAH lesions that showed significantly higher levels of DNA methylation in seven loci: 2C35, CDKN2A ex2, CDX2, HOXA1, NEUROD1, TMEFF2 and TWIST1. Whether this elevated DNA is somehow related to the propensity to progress will require further studies, but it is notable that four of these loci are ones that were designated “intermediate” for increased DNA methylation in AIS. The gene discussed is TMEFF2; the disease is androgen insensitivity syndrome.
