In this study, we tested five assays, mainly using HT1080 fibrosarcoma cell line and its derivatives: (i) luciferase reporter assay (to assess the effects on the Reck promoter in human cells), (ii) immunoblot assay with several human tumor cell lines (to see whether endogenous RECK protein can be up-regulated), (iii) gelatin zymography with culture supernatant (to assess the effects on gelatinases), (iv) time-lapse microscopy (to assess the effects on cell morphology and motility), and (v) subcutaneous inoculation into nude mice (to assess the effects on tumor growth and metastasis).
