In the controlled clinical studies as both monotherapy and combination therapy with metformin\pioglitazone the overall adverse reactions, hypoglycemia, and discontinuation of therapy due to clinical adverse reactions with sitagliptin were similar to placebo and there were no statistically significant differences in the incidence of either hypoglycemia or predefined gastrointestinal adverse experiences between sitagliptin and placebo groups.[4, 5] Due to the glucose-dependent nature of the incretin hormones, insulinotrophic effects do not occur during euglycemia or hypoglycemia.
