We verified CCNB1 mRNA down-regulation following U0126-treatment independently in all colon carcinoma cell lines (Figure 5A) and determined CCNB1 promoter activity in transiently transfected HCT116 cells using a chloramphenicol acetyltransferase (CAT) reporter gene controlled by the 240-bp promoter fragment harboring the two NFY-binding sites [32]. The gene discussed is CAT; the disease is colon carcinoma.
