Because binding of the fVII to TF could induce disseminated intravascular coagulation, a potentially lethal vascular disease, an amino-acid substitution was introduced into the fVII domain of the hI-con1 (Lysine 341 to Alanine) to inhibit initiation of the coagulation pathway without reducing the strong affinity for TF (Hu et al, 1999; Dickinson et al, 2006).
